Abstract:

Fluorescence in situ hybridization (FISH) is a combination of non radioactive molecular biology and cytogenetics. It has many advantages, such as safe use, short experimental cycle, high accuracy and convenient observation. It is widely used in the diagnosis of prenatal, tumor and blood diseases. The process of this technology is complex and tedious, the reagent consumables required are extremely expensive, the professional quality of operators is extremely dependent, and the cost of technical training is high, which greatly reduces the effectiveness of the technology and greatly limits the use and promotion of the technology. In view of the development status of fluorescence in situ hybridization (FISH) and its automation, a fully automatic fluorescence in situ hybridization (FISH) system with high automation, low reagent consumption and high processing efficiency was researched and developed in this paper. In this paper, based on the microfluidic chip technology and medical molecular diagnosis technology with less reagent consumption, fast detection speed, simple operation, multi-functional integration and small reaction volume, the full-automatic liquid-based cell fluorescence in situ hybridization system was studied, which integrated the separation, purification, fixation and hybridization of liquid-based cells It can not only reduce the volume of equipment, but also ensure the sample processing capacity. The processing efficiency is significantly improved. At the same time, the cost and complexity can be greatly reduced, and the amount of reagent that can be controlled is more accurate.
