3D panoramic imaging for cytopatological analysis in light microscopic system

Three dimensional panoramic imaging is an essential tecnique for expanding narrow field of view in microscopic systems. Pathologists can analyze on three dimensional images with wide field of view using this technique. In general, microscopes (confocal, stereo, electron) were used which give the three-dimensional structure of the sample and extra algorithms were not used to extract the three dimensional structure in the literature studies proposed for the development of three dimensional panoramic imaging technique. In contrast to studies in the literature, the specimen prepared for cytopathological analyses was scanned with a light microscope and the three dimensional structure of the sample with wide field of view was obtained in this study. The proposed study consists of two main steps: extraction of three dimensional images and three dimensional panoramic imaging. The data set used in the study was obtained by scanning the specimen prepared for pleural effusion for cytopathological analyses with 20X magnification objective. In addition, a motorized system has been developed that allows automatic focusing and scanning by moving in X-Y-Z axes to control microscope.
