The influence of the composition of aluminum- silica with lithium salt or melatonin of endothelial cell line EA.Hy926 function and survival in vitro

This work described endothelial cell line EA.Hy926 viability under composition of lithium salt or melatonin and aluminum oxide with polydimethylsiloxane (Li/ Al/PDMS or Mt/ AI/PDMS) in vitro. Cytotoxicity of the Li/AI/PDMS or Mt/Al/PDMS on EA.Hy926 cells were studied using MTT test and vital dye staining, and NO levels in conditioned media were studied. It was found that AI/PDMS, Li/AL/PDMS and Mt/AI/PDMS significantly decreased survival rate and increased secretory potential of EA.Hy926 cells in vitro. So, AI/PDMS, Li/AL/PDMS and Mt/AI/PDMS significantly decreased NAD (P) H-dependent oxidoreductase intracellular enzyme activity of EA.Hy926 cells compared with control (p≤0.05). Also, AI/PDMS, Li/AL/PDMS and Mt/ Al/PDMS significantly increased NO production by EA.Hy926 cells compared with control (p≤0.05). Using acridine orange/ethidium bromide dye was estimated increased rate of apoptosis/necrosis of EA.Hy926 cell under AI/PDMS, Li/AL/PDMS and Mt/AI/PDMS condition (p≤0.05). While, weakly induced EA.Hy926 cells apoptosis/necrosis by Mt/Al/PDMS was observed.
