Association of inflammation, destruction and angiogenesis genes polymorphism with serum levels of matrix metalloproteinases 3 and 9 in patients with coronary atherosclerosis

matrix metalloproteinases play a significant role in atherosclerosis, while they are actively regulated by cytokines and growth factors. The aim: analyze the frequency of distribution of promoter genes combinations of cytokines TNFa, IL1β, IL2, IL4, IL6, IL10, regulatory sites of vascular endothelial growth factor VEGF, genes of matrix metalloproteinases MMP2, MMP3, MMP9 with serum levels of MMP3 and MMP9 in patients with coronary atherosclerosis. Materials and methods: The study included 83 men with coronary atherosclerosis. MMP3 and MMP9 serum level were determined by enzyme immunoassay. Polymorphism studies were carried out by RT-PCR using SYBR GreenI and TaqMan probe method. The statistical analysis used a quantile approach, the criterion χ2 with the Yates correction and two-sided Fisher exact test. Results: Complex genetic combinations which positively associated with high MMR3 level were identified. Presumably, the combination of two polymorphic positions of IL-10-592C:MMR3 -1171 5A6A is the leading one. 26 complex genotypes were associated with low serum MMP3 levels. Eight genetic combinations are associated with high MMP9 production. At the same time, there were no associations of the analyzed genotypes with a low level of MMP9 production. Conclusion: the data obtained by us confirm the complexity of the mechanisms for regulating the production of matrix metalloproteinases in atherosclerotic processes and emphasize the need and importance of understanding complex gene interactions in the analysis of certain processes
