A Developed Micro Electric Impedance Spectroscopy for Biological Cells’ Electrical Parameters Extraction

Lab-on-a-chip (LOC) and microfluidic devices, such as micro electric impedance spectroscopy (µEIS), are very tiny devices that use fluid as a working medium in conjunction with biological cells to obtain different dielectric parameters of the used biological cells. Many advantages can be provided by using these tiny microfluidic devices, such as portability, disposable, and high accuracy. Dielectric parameters of biological cells are one of the interesting parameters that can be extracted using µEIS. In this paper, a developed µEIS device is presented and experimentally used to obtain and monitor the change of the dielectric features of U2OS cells defined as Human Bone Osteosarcoma Epithelial Cells with time. Also, the developed µEIS is used to obtain the dielectric spectroscopy features of U2OS cells (i.e., the change of the dielectric parameters with frequency). The used frequency range is from 1K Hz to 3M Hz at room temperature and the sample volume is 10µL. The experiment is conducted for 24 hours and the impedance change is monitored and recorded. An impedance measurement comparison with the dead U2OS cells is provided where a difference can be noticed.
