Potential Polymorphism of BMP-2 rs235768 in North Sumatra sub-population with Mandibular Asymmetry

The potent Bone Morphogenetic Protein-2 (BMP- 2) involved in bone remodeling during post-natal bone growth, such as osteoarthritis and osteoporosis. The expression of BMP- 2 in the proliferative chondrocyte of condylar hyperplasia cartilage effected the mandibular growth and development. This paper aims to analyze the single-nucleotide polymorphism (SNPs) of BMP-2 c.893 A>T (rs235768) in North Sumatra subpopulation orthodontic patients with mandibular asymmetry. A cross-sectional study in fifty-four orthodontic patients with mandibular asymmetry based on Lemos asymmetry analysis. The DNA extraction from peripheral blood followed with amplification and restriction enzyme digestion procedure in polymorphism analysis. There were minor alleles AT (25.9%) and TT (74.1%) in BMP-2 rs235768 were detected whilst the other minor allele (AA) none in this studied subjects with mean age 21.62 to 2.36 years old. BMP-2 protein levels of those patients were measured via enzyme- linked immunosorbent essay (ELISA) method showed the serum level for AT 40.99 to 13.73 ng/dl and for TT 32.23 to 7.09 ng/dl. This baseline study might be an important issue to understand the multifactorial of mandibular asymmetry and temporomandibular disorder that might be related to osteoarthritis and osteoporosis.
