Nanocomposites Polymer/Gold Nanoparticles/Chlorin e6 for Antitumor Laser Medicine

In the present study we have focused on synthesis and characterization of nanocomposite for photodynamic therapy. The nanocomposite consisted of gold nanoparticles and photosensitizer Chlorin e6 incorporated into star-like polymer Dextran-graft-Polyacrylamide. The behavior of three-component nanosystem in the region of physiological temperatures was studied by using Dynamic Light Scattering. A small change in the size of polymer aggregates was observed at an increase in temperature from 25 ° C to 45 ° C. However, size of gold nanoparticles remained around 10 nm at all temperatures. The absorption spectra of nanosystem revealed the interaction of the photosensitizer Chlorine e6 with gold nanoparticles. In vitro testing of studied nanocomposite efficiency for Photodynamic therapy was carried out on the sub-lines of breast carcinoma MCF-7/S; MCF-7/Dox, and Jurkat cells. After irradiation with laser the death of 68.4% of MCF-7/S cells was registered. However, for MCF-7/Dox cells - 24.9%, for Jurkat cell line - 20 %. It was proved that prhotodynamic efficacy of proposed nanocomposite was depended on malignant cells type, while slight aggregation process not significantly affected nanosystem activity.
