Effect of Potassium Iodide Use on Anti-Cancer PDT Applications on PC-3 Prostate Cancer Cells

Photodynamic therapy (PDT) is a promising alternative method in the treatment of cancer. The mechanism of PDT is based on the production of reactive oxygen species (ROS) as a result of the interaction of the main components of molecular oxygen, light and photosensitizer. Therapeutic approaches can be achieved through these interactions. In cases where the effect of PDT is insufficient, various agents are being investigated to increase the effect. In this study, Potassium Iodide (KI) was used as an enhancer to increase the anticancer activity of PDT in the PC-3 cell line. The light source was diode laser at 655 nm wavelength and its output power was 200 mW, and energy doses were 0.5, 5, 10 and 20 J/cm
²
. Different concentrations of toluidine blue ortho (TBO) (0.5 ¼M, 5 ¼M, 50 ¼M, and 100 ¼M) were used as a photosensitizer. Cell viability was assessed by MTT analysis after applications. The most effective result in a combination of PDT with KI was obtained as a 78% reduction in cell viability. However, this result does not support the development of photodynamic therapy applications, because similar cancer cell death rates were obtained without the use of KI.
