Development of Peptide Aptamers – 3d Complementary Ligands for Selective Binding of Target Biomarkers of Diseases in Multiparametric Sensor Systems

Biosensor system based on 3D peptide-protein molecular recognition mechanism for rapid medical diagnostics of diseases by determination their protein biomarkers in biological fluids has been considered. The diagnostic system was implemented as micro-arrays with topologically structured sites of bonded peptide aptamers (PMA). The perspective 3D structures of peptide aptamers were analyzed using the Conjugated Ion-Hydrogen Bonds System (CIHBS) developed earlier. A number of peptide aptamers with spartial complemetarity to myoglobin and troponin were designed, synthesized and tested as ligands for myocardial infarction diagnostics biochip. The testing of aptamers affinity to target biomarkers were investigated using capillary electrophoresis-on-a-chip with an Experion Bio-Rad Pro260 system. This method enables a complex formation of peptide and target protein to be analyzed. As a result three aptamers were proved to be complementary to target proteins: Troponin I and Myoglobin. A topology and design of microfluidic diagnostic biochip has been discussed.
