Development of Heterologous Cell Co-culture Technique for Application to Bioreactor

In this research, as a technique to realize an eco-friendly society, heterologous cell micropatterning technique is developed for controlling the cell surrounding environment of bioreactor. And we evaluate that the effect of cell cultured environment to cell substance production effect. The cell micropattern was created by general micromachining method similar technique such as photolithography and wet etching. This technique can create the precisely micropattern of single cell level. Also, a cell micropattern consist of different types of cells was successfully created using the presented technique. After the creation of the heterologous cell micropattern, a simple 3dimensional tissue (spheroid) was disseminated and co-cultured on the created cell micropattern. Then, cell culture supernatants were collected by micropipette, and were evaluated the cell substance production ability. As a result of the cell substance production ability, spheroids co-cultured on the heterologous cells micropattern was significantly higher production ability than other conditions. The production ability of the spheroid cultured on the heterologous cells were achieved 300% production compared with the spheroids cultured on the non patterned heterologous cells. This results indicate that the heterologous cells patterning and co-culture technique are useful for the development of effective bioreactor.
