Nspefs Promoting the Proliferation of Piec Cells: an in Vitro Study

Currently, nanosecond pulsed electric fields(nsPEFs) with short pulse duration, low energy density and nonthermal effects have many practical applications in medicine and biology, especially in tumor ablation. But there are a few researches in its proliferation in normal cell. Proliferation of endothelial cells can accelerate the stent endothelialization and reduce the incidence of acute thrombosis. In this current study, we evaluated the proliferation effect of nsPEFs on porcine iliac endothelial cell line and explored the potential mechanism involved. PIEC cell line was tested by CCK-8 assay for cell proliferation, intracellular Ca

concentration was measured using the fluorescence Ca
²⁺
indicator fluo-4 AM, reactive oxygen species assay for intracellular reactive oxygen species (ROS) and total nitric oxide assay for NO production in the cultured medium. Results showed that low field strength (100ns, 5kV/cm, 10Pulses) had 16% of the proliferation on PIEC cells after 48 hours' post-treatment. The intracellular Ca
²⁺
concentration and ROS were increased after nsPEFs treatment. Additionally, NO concentration also increased after nsPEFs treatment. Our results demonstrate that appropriate pulse parameters can enhance cell proliferation on PIEC cells and this may involve the intracellular Ca
²⁺
concertation, ROS and NO production in nsPEFs treatment system. These results suggest that nsPEFs may have a potential application in the process of accelerating the stent endothelialization.
