Human Papilloma Virus biosensor based on electrochemical impedance spectroscopy of DNA hybridization

The aim of this study is to perfect detection of a high risk type of Human Papilloma Virus (HPV) DNA. HPV infection is the most common risk factor and causative agent of cervical cancer. HPV DNA testing is widely used in cervical cancer screening programs to improve the detection of high-grade lesions. Herein we report a label-free electrochemical impedance sensor for recognition of the HPV DNA hybridization. Single-stranded 25-base DNA was immobilized at its 5'end onto gold electrode surfaces. After hybridization with complementary or non complementary DNA, electrochemical impedance spectra were obtained using [Fe(CN)6]
^3-/4-
as redox marker ions. Hybridization with the complementary DNA will be resulted in increasing the charge-transfer resistance (R
_CT
), whereas non complementary attachment doesn't have any effect on the R
_CT
. The proposed sensing methodology represents a very attraction alternative to the existing method for the detection of HPV.
