A luminometer for determination of multiple metabolites in undiluted human plasma

A luminescence-based biosensor allowing for multiplex measurement of metabolites in undiluted human plasma is revealed in this study. Concurrent measurement of renal function-related metabolites including creatinine, urea, uric acid and glucose in a complex sample can be achieved in a single step using specially designed compositions which were pre-distributed into the analytical cartridge and freeze-dried into reagent-free powder. The dry-reagents allow for a rapid screening for a small amount of sample down to 10 µL. The total analysis time and required sample volume for an on-cartridge measurement of four analytes are 10 min and 150µL, respectively. We demonstrated that the system provides a good sensitivity and a wide dynamic range for metabolites measurements such that it properly meets the clinical requirements. Evaluation of long-term stability was conducted to verify the shelf life and potential applicability of the developed compositions.
