Development of glucose biosensor based on enzyme immobilization

Experimental investigations have been carried out on the development of a glucose biosensor for monitoring blood plasma glucose, based on specific enzyme immobilization (glucose oxidase-GOD) on a membrane surface coupled to an appropriate transducer. Modified polysulfone (PS) and PVA-PAA blend membranes with GOD immobilization have been developed, under optimal conditions. Enzyme immobilized membrane systems have been characterised with respect to specific activity, selectivity, specificity (with mixture of glucose monomers, lactose, manose, and fructose), stability, pH and temperature optima, reusability, and shelf life with encouraging results for PVA-PAA blend membrane systems within the blood plasma glucose concentrations expected in IDD patients. Kinetic constants (k/sub m/ and v/sub max/) have been improved considerably by GOD immobilization in PVA-PAA with the bis-acrylamide membrane system.
