Fabrication of Replicated DNA Microarray using Polyacrylamide Gel

This paper reports a new DNA chip printing method based on a master DNA chip. In our DNA nanoprinting, a microarray of covalently immobilized single-stranded DNA, a master-chip, is hybridized in mixture of single-stranded DNA molecules solution, which has all complementary sequence of the attached DNA on the master-chip and some acrylamide group at 5' end of DNA. Due to the sequence specific hybridization, hybridized DNA molecules are localized on the master-chip according to their sequences. To make print-chip, acrylamide group attached DNA make polymerization onto a new substrate plate. Polyacrylamide gel is formed between DNA and a printed chip plate. During the gel formation, the acrylamide group of hybridized DNA is copolymerized with acrylamide in solution. And then, the print-chip is detached from the master-chip with DNA incorporated polyacrylamide gel layer. As covalently attached DNA is remaining on the master-chip, which can be reused. Also using this technique, same copy of DNA microarrays can be fabricated repeatedly 10 times. A personalized DNA microarray, which can be generated with similar method, will be also discussed.
