LIFU Accelerates CSF Circulation and Alleviates Motor-Cognitive Dysfunction in Rat with Hydrocephalus

Hydrocephalus is a neurological disorder characterized by abnormal accumulation of cerebrospinal fluid (CSF), leading to ventricular enlargement and motor-cognitive dysfunction. While shunt surgery remains the standard treatment, it carries risks of complications. This study aimed to evaluate the therapeutic effects of low-intensity focused ultrasound (LIFU) on CSF circulation and motor-cognitive function in a rat model of hydrocephalus. Adult Sprague-Dawley rats were randomly assigned to three groups: Control group (no treatment), Hydro group (hydrocephalus model), and Hydro+LIFU group (hydrocephalus model treated with LIFU). LIFU was applied daily to the third ventricle for 21 days. Behavioral tests, body weight assessment, magnetic resonance imaging (MRI), and in vivo imaging were performed to assess the therapeutic effects. Compared to Hydro group, Hydro+LIFU group exhibited significant improvements in body weight, motor-cognitive performance, ventricular size, and CSF circulation. These results suggest that LIFU could be a promising, non-invasive treatment for hydrocephalus with significant potential for translation into clinical practice.
