Electrokinetic-Based Micro-Nanofluidic Device for Sarcoma Extracellular Vesicle Isolation for Rapid Point-of-Care Detection

This paper reports on a modular, hybrid micro-nanofluidic device that exploits electrokinetics while integrating point-of-care technology advances. Our device combines size-based isolation using a nanofluidic-membrane with immunoaffinity-based capture and engineered surface coatings, achieving a 4.2-fold increase in isolated extracellular vesicles (EVs), a 5-fold increase in EV-DNA/mL of biofluid, and a 75% reduction in isolation time compared to ultracentrifugation (UC). These advances show significant promise in liquid-biopsy-based point-of-care devices for liposarcoma detection.
