Abstract:

To achieve efficient messenger ribonucleic acid (mRNA) extraction during transcription–translation coupled with association of puromycin linker (TRAP) display screening process, we reported a new microfluidic chip to perform mRNA extraction from in vitro transcribed reaction mixture without high-speed centrifugation, solvents, and cooling conditions by using mRNA-probe-coated magnetic beads, which could automate the entire In vitro transcription (IVT), DNase treatment and mRNA extraction process on a single chip. It is critical for on-chip TRAP display screening. With this approach, puromycin-linked mRNA products, which can be utilized in peptide screening to target KIF2C, an important protein overexpressed in certain types of cancer, including triple-negative breast cancer (TNBC), may be extracted with a high yield.
