Investigating Photosensitizing Properties of Methylene Blue on Perfusate Solutions Decontamination for Organ Preservation

In this study, methylene blue (MB) was evaluated as a photosensitizer for photodynamic therapy aimed for decontaminating organ perfusates used during transplantation. The absorption spectroscopy and liquid chromatography coupled to mass spectrometry (LC-MS) were employed to follow up the MB photodegradation. The in vitro antimicrobial efficacy of photodynamic therapy was investigated through microbiological assays using Staphylococcus aureus. The findings suggest the interaction of MB with biomolecules present in perfusates significantly attenuate its photodynamic activity, potentially by trapping its molecules and thereby diminishing its interaction with bacterial cells.
