Possibilities of Holotomographic Microscopy for Studying Primary Cell Cultures of the Brain

Holotomography is one of the types of microscopy of the twenty-first century. The possibility of long-term in vitro imaging of cells is realized in many imagers capable of imaging using light microscopy, as well as in confocal and fluorescent microscopes with an incubation chamber. Using the example of neuronal and glial primary cell cultures, we will demonstrate the advantages and situations in which holotomographic microscopy is irreplaceable, and the difficulties that the researcher faces when working with this imaging method. In particular, high resolution with the ability to visualize the thinnest processes such as axons, mitochondria inside them, the structure of the nucleolus is combined with long mathematical calculations, and the lack of free software.
