Synthesis of Ultrasound Sensitive Bovine Serum Albumin Nanosystem for On Demand Release of Camptothecin in Triple Negative Breast Cancer Cell lines

Ultrasound stands as a pivotal therapy for cancer, utilizing its non-invasive nature to enable targeted treatments such as enhanced drug delivery, and precise modulation of cellular responses. The development of an ultrasonic-sensitive nanosystem capable of effectively targeting cancer and preventing its spread or recurrence is highly desired. This study focuses on the synthesis of an innovative ultrasound-sensitive nanosystem using Bovine Serum Albumin (BSA) as a carrier for the controlled release of Camptothecin (CPT) in Triple Negative Breast Cancer (TNBC) cell lines. The nanosystem is designed to respond to ultrasound stimuli, enabling on-demand drug release. The formulation process involves the self-assembly of BSA and CPT, forming a stable and biocompatible nanocarrier. Ultrasound sensitivity is achieved through the incorporation of Tartrazine. In vitro studies showed the high therapeutic efficiency of CPT@BSA NPs, including efficient cellular absorption, a high percentage of cell death (70-80 %), reduction of cell proliferation after treatment, and high cell death in 3D spheroids.
