Identification between tea-cigarette and cigarette based on the Nicotiana-Specific DNA molecular marker

Exploration of efficient and accurate identification between tea tea-cigarette products and tra traditional cigarettes is of great importance to define and supervise the market which are essential to ensure the rights and interests of consumers. In this study, 21 tea tea-cigarette products that are widely marketed were used as materials. With tobacco raw ma materials as positive control, and non non-tobacco raw materials as negative control, NicotianaNicotiana-specific molecular markers (Ntsp027 and Ntsp151) were used to identify and test the tea tea-cigarette products collected. Meanwhile, the nicotine content in the smoke of the tested tea tea-cigarette products was also detected. The results showed that 2 of the 21 tea tea-cigarette products M20 and M21, amplified tobacco genus genus-specific bands (i.e., 303 bp and 300 bp tobacco genus genus-specific fragments were amplified by Ntsp027 and Ntsp Ntsp151, respectively) after testing with tobacco genus genus-specific molecular markers, indicating that tea tea-cigarette products M20 and M21 contained tobacco raw materials. The GC GC-MS detection found that the smoke produced by burning 3 products M04, M20 and M21 con contained nicotine, and their nicotine contents were 0.92 mg/cig., 0.94 mg/cig. and 0.83 mg/cig., respectively, demonstrating that the 3 products either contained nicotinic raw materials or exogenously added nicotine. Based on the two collaborative detection methods of tobacco Nicotiana-specific markers and GC GC-MS, they can accurately determine that the two commercially available tea tea-cigarettes of M20 and M21 tested contain tobacco raw materials, which belong to the category of cigarettes and should be included in tobacco monopoly supervision; while the tea tea-cigarette product M04 tested adds exogenous nicotine components. The above results further show that the tobacco Nicotiana specific DNA marker invented is of great efficiency and precision for the identificat identification of tobacco raw material involvement, which is a credible supplement for the current GC GC-MS strategy of merely considering the nicotine as the category evidence of tobacco raw materials.
