Modeling of the DNA Melting Point Dependence on Various Analysis Factors

This article describes the factors influencing the value of the DNA melting temperature $T_{m}$. There was revealed the necessity of creating new models that would allow more accurate $T_{m}$ determination when some factors were changed. The ANK-32 nucleic acid analyzer was used for analysis by the melting method of three known DNA samples. As a result, the error of the average melting temperature was estimated: the RMS values for all samples in 4 test tubes are no more than 0,04 °C. To reduce the error a refined model was proposed for calculating the melting temperature of DNA, taking into account the dependence on the length of the DNA fragment and the content of GC-base pairs. It is shown that when using the refined model the melting point calculation deviation from the experimental data does not exceed 0,003 °C. The model provides an opportunity to compare the results of experimental determination of $T_{m}$ values with theoretically predicted values.
