Effect of combined tera- and gigahertz irradiation on hemolytic features of human erythrocytes in vitro

The purpose of this work was an examination of the effect of combined low-intensive terahertz (THz) (f = 0,89 THz, P = 1,5 mW/cm²) and gigahertz (GHz) (f = 37,7 GHz, P = 2,7 mW/cm²) irradiation on osmotic resistance of human erythrocytes in vitro. The evaluation of erythrocytes hemolysis by a concentration of peroxide oxidation of lipids (POL) products such as malonic dialdehyde (MDA) and diene dialdehydes (DD) was carried out. The erythrocytes hemolysis factor at the presence of hypotonic salt solutions NaCl was estimated. The result of combined action of electromagnetic waves THz and GHz rages on peroxidation of lipide phase and erythrocytes anti-oxidant status was given. It is shown probably an anti-oxidant effect of low-energy laser irradiation under conditions of pulse modulation with frequencies f = 200 ÷ 450 Hz. Subject of investigation was the human venous blood (middle age - 40 years). The blood was centrifuged softly under condition less than 1000 rot./m. up to complete washing-out cells in 0,9% solution NaCl with heparinum. Thus the hematocrit was 94 - 96%. The stability of erythrocytes membrane by the method of definition of an osmotic resistance was detected. Also hemolysis percent and intensity of POL were estimated. The hemolysis parameter was calculated by formula Ex x 100 / El, where El - supernatant extinction in the test-tube with 0,1% NaCl solution, Ex - extinction of investigated sample, 100 - complete erythrocytes hemolysis in the test-tube with 0,1 % NaCl solution. Concentration of products POL - MDA and DD had been defined by reaction with thiobarbituric acid under general technique. The equipment permitting in real time to measure the electromagnetic response of erythrocytes in range of a dielectric dispersion of free water at various expositions of laser irradiation was used [1 - 3]. Terahertz and gigahertz irradiation was modulated by amplitude, at that the laser irradiation was introduced in 8-mm waveguide with help of specially designed for these purposes waveguide cuvette. Data analysis was realized with help of the programs of accumulation of an electromagnetic signal in a millimeter range of radiowaves from under noise and further processing with application of FFT (Fast Fourier Trans formation).
