Dynamic Morphological and Growth Changes in Fibroblasts Post-Thawing: A Time-Dependent Study

This study investigates the morphological recovery and growth dynamics of fibroblast cells at various time intervals (24, 48, and 72 hours) post-thawing under magnifications of 10x/0.3. The analysis provides insights into cellular behaviour and morphology changes over time, contributing to understanding the recovery process of fibroblast cells following cryopreservation. Cryopreservation is an essential step in cell culture experiments and post-thaw recovery has a considerable effect on cell culture downstream applications such as tissue engineering and regenerative therapy. To understand how fibroblasts adapted post-thawing, human dermal fibroblasts were plated in DMEM containing 10% fetal bovine serum and 1% penicillin streptomycin and monitored at 24, 48 and 72 hours post thawing. It was progressively observed that cellular adhesion, proliferation and morphological stability increased over time. Quantitative analysis of fibroblast growth demonstrated a progressive increase in average cell length from 296.8 μm at 24 hours to 390.9 μm at 72 hours which is indicative of a slow tissue remodelling and integration. Cellular stress responses such as membrane blebbing were visible at high magnifications for an early time point, after which the cells resumed their spindle-shaped morphology. These results highlight the critical role of post-thaw conditions in maximizing fibroblast recover. Observations from this study may aid in optimizing post-thawing protocols to maintain cell viability and functionality. Additionally, this study aims to assess changes in the growth of fibroblasts, which will serve as a foundation for further experiments involving this type of cell.
