Application of Principal Component Analysis (PCA) in Bioinformatics Analysis of Osteonecrosis of the Femoral Head (ONFH)

Osteonecrosis of the femoral head (ONFH) is a complex polygenic disorder caused by the combined effects of numerous genetic variations rather than a single gene mutation. Research in this area is crucial for understanding disease mechanisms. In this study, principal component analysis (PCA) was employed for bioinformatics analysis of ONFH. The analysis utilized the GSE123568 gene expression microarray dataset, which consists of 40 samples (10 control and 30 disease). The distribution of gene expression levels across samples was calculated and illustrated on the first two principal components. Differentially expressed genes (DEGs), 318 upregulated and 229 downregulated, were identified and visualized in a volcano plot and heatmap. Significant biological processes, cellular components, and molecular functions were identified through Gene Ontology (GO) enrichment analysis. Key pathways were subsequently detected via Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. These findings provide valuable insights into the molecular mechanisms underlying ONFH and highlight potential therapeutic targets for clinical applications.
