SlipChip-Based Microfluidic Device Efficient Sample Preparation and Multiplexed PCR Detection

Screening for high-risk human papillomavirus (HPV) is essential for the control and prevention of cervical cancer. While laboratory-based nucleic acid amplification tests (NAATs) provide precise HPV diagnosis, they generally necessitate expensive equipment, specialized laboratory facilities, and trained personnel. Combining high-quality sample preparation with rapid multiplexed detection offers substantial promise. In this study, we introduce an injection-molded SlipChip, inspired by the combination lock mechanism, that can execute magnetic bead-based nucleic acid extraction through a series of clockwise and counterclockwise rotations. Furthermore, we have incorporated a portable, four-channel real-time quantitative PCR device for the amplification and detection processes. This system successfully demonstrates the concurrent detection of four PCR channels for HPV screening. This innovation signifies a major step forward in HPV screening, especially for resource-limited environments, and it holds potential for broader applications in NAAT for point-of-care and home-based testing.
