We have developed a new method to measure the temperature distribution in water with high spatial resolution. The temperature distribution was successfully calculated from the fluorescent intensity distribution of the Rhodamin B solution on the micro-thermo device for single molecular observation. With this method, the direct measurement of local temperature, where the molecule of interest stands, can be achieved with accuracy of 2 °C and 5.3 μm in spatial resolution.
