Temperature distribution measurement on micro-thermo device for single biomolecular observation using fluorescent dye

We have developed a new method to measure the temperature distribution in water with high spatial resolution. The temperature distribution was successfully calculated from the fluorescent intensity distribution of the Rhodamin B solution on the micro-thermo device for single molecular observation. With this method, the direct measurement of local temperature, where the molecule of interest stands, can be achieved with accuracy of 2 /spl deg/C and 5.3 /spl mu/m in spatial resolution.
