Engineering of solution-based localized surface plasmon resonance platform for dengue diagnosis

Summary form only given. Dengue virus infection, an arthropod-borne virus transmitted by Aedesaegypti mosquitos, has severe health risks in tropical and sub-tropical areas worldwide [1, 2]. Specific test for Dengue infections are rarely performed, limiting the establishment of an appropriate therapy. Many challenges on fighting Dengue infection still need to be addressed, including the development of reliable, label-free and cost-effective diagnosis techniques. In this article, we present a new optical platform to assay dengue virus in the acute phase of infection.Here we propose a new bios ensor platform based on Localized Surface Plasmon Resonance (LSPR) aqueous solution. FEM calculations were used to investigate the electric field behavior around the nanostructure, and therefor to determine the proper NP dimensions for molecular sensing application. Fig. 1a shows the 2-dimension plasmon field enhancement distribution for the gold LSPR dipole of gold NP with different diameters, indicating that Au NP can enhance the electrical field even at distance greater than 30 nm away from the NP surface. On the development of the sensor platform, Dengue anti-NS1 antibody was immobilized on AuNPs surface. Cysteine molecules were used as ligand in the antibodies functionalization process and glycine as blockers of non-functionalized sites The sensing procedure was base on the evaluation of the LSPR spectra shift, induced attachment to the surface of NPs. Spectral shift was observed after every step of platform [ 3], on gold nanoparticles (NP) in of the platform. by molecular manufacturing of Dengue NS1 antigen, as presented on Fig. 1b.
